Two structural features account for the prolonged half-life of semaglutide. A modified amino acid at position 8 resists cleavage by dipeptidyl peptidase-4, the enzyme that rapidly degrades native GLP-1. A fatty diacid side chain binds serum albumin, which limits renal clearance and protects the peptide from enzymatic breakdown. These modifications yield a plasma half-life of approximately one week in humans, allowing once-weekly administration. The relationship between plasma concentration and clinical effect varies between individuals, and sources of that variability are still being characterized.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1 (GLP-1), a hormone released from intestinal L-cells after food intake. The compound belongs to the incretin mimetic class and acts at GLP-1 receptors distributed across pancreatic, gastrointestinal, cardiovascular, and central nervous system tissues. Compared with native GLP-1, the molecule carries structural changes that extend its activity from minutes to roughly one week. It is studied for glycemic control in type 2 diabetes and for weight management, and its effects on cardiovascular and other outcomes remain active research areas.
Receptor binding triggers G protein signaling that raises intracellular cyclic AMP in pancreatic beta cells. Insulin release follows in a glucose-dependent manner, so secretion increases when blood glucose is elevated and diminishes when it is not. The same signaling suppresses glucagon release from alpha cells and slows gastric emptying, which blunts the post-meal glucose rise. In the brain, receptor activation in regions such as the arcuate nucleus is associated with reduced appetite and lower energy intake. How much each of these effects contributes to overall weight change is not fully settled.

Lyophilized semaglutide powder is typically held at minus twenty degrees Celsius for long-term storage. At that temperature, solid-state degradation reactions proceed slowly and the peptide remains intact for extended periods. Repeated freeze-thaw cycles are best avoided because they promote aggregation and can shift the proportion of monomeric peptide present. Working aliquots are often prepared so that each portion is thawed only once, and desiccant is placed inside the vial to limit moisture uptake.
Identity and purity are usually assessed by reverse-phase high-performance liquid chromatography coupled to mass spectrometry. Retention time and observed mass are compared against a reference standard run under identical conditions. Impurity profiles reveal deamidation products, oxidized methionine variants, and truncated fragments that arise during synthesis or storage. Peptide mapping through enzymatic digestion confirms the primary sequence, while amino acid analysis offers an independent check on overall composition.
Routine characterisation of the peptide relies on reversed-phase high-performance liquid chromatography, often paired with ultraviolet detection near 214 nanometres. Related substances such as deamidated, oxidised, and truncated sequences elute at characteristic positions and are quantified by area percentage. Electrospray ionisation mass spectrometry confirms the molecular mass and can resolve some closely related variants. Peptide mapping after enzymatic digestion provides sequence-level verification and is useful when a full identity profile is required. Method parameters such as column chemistry, gradient, and mobile-phase pH influence the separation and must be reported alongside results.
Material described as research-grade is not necessarily manufactured to pharmaceutical standards, and purity figures depend on the method used to obtain them. A certificate of analysis states the measured purity, the analytical technique, and the batch identifier, but the underlying data are not always included. Independent testing by a second laboratory is a common way to confirm identity and purity. Uncertainties remain about how storage history affects long-term stability, and about how well results from one laboratory transfer to another. Documentation of handling conditions supports comparison between batches.
Peptides are sensitive to temperature, light, oxygen, and repeated freeze-thaw cycles. Semaglutide in dry form is generally held at refrigerated temperatures, while reconstituted solutions require a defined short-term storage window. Vials should be kept in secondary packaging to limit photodegradation, and exposure to alkaline conditions is avoided because it accelerates chemical degradation. Adsorption to glass and some plastics can reduce the measured concentration of dilute solutions, so low-binding polypropylene containers are preferred for analytical work. Each transfer step introduces a small risk of contamination, and closed handling practices reduce that risk.
Prostaglandin E1 (PGE1) is a naturally occurring prostaglandin with various medical uses. Alprostadil and misoprostol are synthetic forms of prostaglandin E1 used as medications. Lubiprostone, a derivative of prostaglandin E1, is also used as a medication. Prostaglandin E1 is a vasodilator. It has various effects in the body that include opening blood vessels, relaxing smooth muscle, inhibiting clotting, and causing uterine contractions. In infants with certain congenital heart defects, alprostadil is delivered by slow injection into a vein to maintain a patent ductus arteriosus until surgery can be carried out. By injection into the penis or placement in the urethra, alprostadil is used to treat erectile dysfunction. Common side effects when given to babies include decreased breathing, fever, and low blood pressure. When injected into the penis for erectile dysfunction; side effects may include penile pain, bleeding at the site of injection, and prolonged erection (priapism). Prostaglandin E1 was isolated in 1957 and approved for medical use in the United States in 1981. Misoprostol has various obstetric uses. It is used to induce abortion, to completely empty the uterus after a miscarriage, to induce labor, and to prevent and treat postpartum hemorrhage. The medication is available through many routes. It can be swallowed, dissolved in the mouth, placed in the vagina, or placed in the rectum. Misoprostol can also be used to manage duodenal ulcers and peptic ulcer disease when other medications are not effective.
=== 1945 === February 4–11: The Yalta Conference in Crimea, RSFSR, with US President Franklin D. Roosevelt, British Prime Minister Winston Churchill and Soviet leader Joseph Stalin, and their top aides. Main attention is deciding the post-war status of Germany. The Allies of World War II (the United States, the Soviet Union, United Kingdom and also France) divide Germany into four occupation zones. The Allied nations agree that free elections are to be held in Poland and all countries occupied by Nazi Germany. In addition, the new United Nations are to replace the failed League of Nations. March 6: The Soviet Union installs a puppet government in Romania. March 7: Josip Broz Tito is installed as the head of the provisional government of Democratic Federal Yugoslavia. March–April: U.S. and Britain outraged as Stalin excludes them from a role in Poland and turns Poland over to a Communist puppet government he controls. March–April: Stalin is outraged at inaccurate reports about Operation Sunrise that American Office of Strategic Services in Switzerland is negotiating a surrender of German forces; he demands a Soviet general be present at all negotiations. Roosevelt vehemently denies the allegation but closes down the operation in Switzerland. A Soviet general is present at the negotiations in northern Italy that lead to surrender. April 12: Roosevelt dies; Vice President Harry S. Truman takes over with little knowledge of current diplomatic efforts, no knowledge of the atomic bomb, and a bias against Russia. April 28: Benito Mussolini dies. April 30: Adolf Hitler dies.
Because the city had flourished in the heyday of the auto industry, local politicians made periodic attempts to stimulate a revival of the auto industry in the city. For example, in the 1980s the cities of Detroit and Hamtramck used the power of eminent domain to level part of what had been Poletown to make a parking lot for a new automobile factory. On that site, a new, low-rise suburban type Cadillac plant was built, with substantial government subsidies. The new Detroit/Hamtramck Assembly employs 1,600 workers. In the 1990s, the city subsidized the building of a new Chrysler plant on the city's east side, Jefferson North Assembly, which employs 4,600 people. In 2009 Chrysler filed a Chapter 11 bankruptcy case, and survives in a partnership with Fiat of Italy, while GM filed for Chapter 11 bankruptcy on June 1, 2009, and survives as a much smaller company—smaller now than Japan's Toyota Motor Corporation. A little over two years after these major blows to the U.S. auto industry, the city itself went into Chapter 9 bankruptcy after years of mismanagement by local leaders.
=== Sperm morphology === Regarding sperm morphology, the WHO criteria as described in 2021 state that a sample is normal (samples from men whose partners had a pregnancy in the last 12 months) if 4% (or 5th centile) or more of the observed sperm have normal morphology. If the sample has less than 4% of morphologically normal spermatozoa, it's classified as teratozoospermia. Normal sperm morphology is hard to classify because of lack of objectivity and variations in interpretation, for instance. In order to classify spermatozoa as normal or abnormal, the different parts should be considered. Sperm has a head, a midpiece and a tail. Firstly, the head should be oval-shaped, smooth and with a regular outline. What is more, the acrosomal region should comprise the 40–70% area of the head, be defined and not contain large vacuoles. The amount of vacuoles should not excess the 20% of the head's area. It should be 4–5 μm long and a width of 2.5–3.5 μm. Secondly, the midpiece and the neck should be regular, with a maximal width of 1 μm and a length of 7–8 μm. The axis of the midpiece should be aligned with the major axis of the head. Finally, the tail should be thinner than the midpiece and have a length of 45 μm approximately and a constant diameter along its length. It is important that it is not rolled up. Since abnormalities are frequently mixed, the teratozoospermia index (TZI) is really helpful. This index is the mean number of abnormalities per abnormal sperm. To calculate it, 200 spermatozoa are counted (this is a good number).

==== Section B: Thermodynamics, Equipment and Systems ==== Section B on Thermodynamics, Equipment and Systems of the IIR focuses on the technological and scientific fundamentals of classical refrigeration, excluding cryogenic temperatures. The fundamentals are represented by its Commission B1 Thermodynamics and Transfer Processes, whereas Commission B2 Refrigerating Equipment covers all kinds of refrigeration technology. Section B is a key player in most of the IIR international conferences; except for the International Conference of Refrigeration (ICR), organised every four years for all 10 IIR Commissions, where approximately 50% of all presentations are related to Section B topics. Independently, and together with other Sections, Section B hosts a multitude of conferences such as the Gustav Lorentzen Conference on Natural Working Fluids and the Ohrid Conference on Ammonia and CO2 Refrigeration Technologies; or conferences on Thermodynamic Properties and Transfer Processes of Refrigerants, on Magnetic Refrigeration at Room Temperature, on Compressors and Coolants, and on Phase Change Materials and Slurries for Refrigeration and Air Conditioning. Several Working Groups, where emerging topics in refrigeration are discussed by IIR experts to publish results in handbooks or other forms of publications, are organised within the scope of Section B.
==== Operation Freedom's Sentinel ==== Following the withdrawal of U.S. troops from Afghanistan at the end of 2014, the War in Afghanistan continued with 12,000 U.S. and NATO troops are deployed in Afghanistan as part of NATOs Resolute Support Mission whose purpose is to train, advise and assist Afghan government forces against anti-government forces and to conduct counter-terrorist missions. US forces in Afghanistan are deployed under Operation Freedom's Sentinel. The Green Berets took part in the Battle of Kunduz, helping Afghan forces retake the city by October 2015 after it fell to Taliban insurgents. Green Berets have also been assisting Afghan forces in Helmand Province, where on the January 5, 2016, during a major operation aimed at reclaiming territory held by the Taliban, Staff Seargent Matthew McClintock of A Company, 1st Battalion, 19th SFG was killed by small arms fire during an hours long battle in the Marjah district. Two other U.S. troops and four Afghan soldiers were injured. The coalition conducted 12 airstrikes in the area with a variety of aircraft, including F-16 fighter jets and an AC-130 gunship. Two HH-60 Pave Hawk medevac helicopters responded to evacuate casualties, one was waved off and left the scene safely, but the second landed and its main rotor blade was damaged when the helicopter hit a wall.
=== Later history === In the 1960s, virologist and cancer researcher Chester M. Southam injected HeLa cancer cells into patients at the Jewish Chronic Disease Hospital. When three doctors resigned after refusing to inject patients without their consent, the experiment gained considerable media attention. The NIH was a major source of funding for Southam's research and required all research involving human subjects to obtain their consent before any experimentation. Upon investigating all of their grantee institutions, the NIH discovered that the majority of them did not protect the rights of human subjects. From then on, the NIH has required all grantee institutions to approve any research proposals involving human experimentation with review boards. In 1967, the Division of Regional Medical Programs was created to administer grants for research for heart disease, cancer, and strokes. That same year, the NIH director lobbied the White House for increased federal funding to increase research and the speed with which health benefits could be brought to the people. An advisory committee was formed to oversee the further development of the NIH and its research programs. By 1971, cancer research was in full force, and President Nixon signed the National Cancer Act, initiating a National Cancer Program, President's Cancer Panel, National Cancer Advisory Board, and 15 new research, training, and demonstration centers. Funding for the NIH has often been a source of contention in the US Congress, serving as a proxy for the political currents of the time.

Native GLP-1 is degraded within minutes by dipeptidyl peptidase-4 and cleared quickly. Semaglutide carries a position 8 substitution that blocks that cleavage and a fatty diacid chain that binds albumin. Together these changes extend its circulating half-life to about one week.
Yes. Stimulation of insulin secretion is glucose-dependent, meaning the effect is larger when blood glucose is high and minimal when it is normal. This property separates GLP-1 receptor agonists from agents that drive insulin release regardless of glucose level.
Receptor expression in hypothalamic and brainstem regions is well documented, and reduced energy intake is consistently observed. The relative contribution of central versus peripheral signaling to total weight change is still an open question addressed by ongoing research.
冻融过程中冰晶形成与局部浓度升高会促使肽链发生界面吸附和聚集。聚集不仅降低有效含量,还会改变可见异物与不溶性微粒的计数结果。将溶液分装为单次使用的小体积等份可减少循环次数。
