This is a working overview of 二肽基肽酶-4, written for readers who want more than a one-paragraph summary but less than a textbook.

The sequence incorporates alpha-aminoisobutyric acid at position 8, replacing the alanine found in the natural hormone. This substitution blocks the primary DPP-4 recognition site and contributes most of the enzymatic stability. Albumin binding further protects the peptide and reduces the frequency of administration required to maintain active plasma levels. Because the fatty acid chain increases lipophilicity, the compound is formulated as a solution rather than a simple aqueous buffer. Researchers describe the design as an incremental optimization of earlier GLP-1 analogs rather than a wholly new scaffold.
Reported molecular weight is approximately 4113.6 daltons for the free base, and the peptide is supplied as a lyophilized powder or in buffered liquid form depending on the intended use. It is freely soluble in water when formulated with appropriate excipients, though the unconjugated peptide shows limited stability at neutral pH over long periods. Analytical characterization typically relies on reversed-phase high-performance liquid chromatography and mass spectrometry. Purity specifications for research-grade material commonly exceed ninety-five percent by area. Isotopic and impurity profiles differ between suppliers.
Two principal therapeutic variants exist under separate regulatory filings, one indicated for glycemic control in type 2 diabetes and one for chronic weight management. Both use the same active molecule; differences lie in formulation strength, titration schedule, and labeling. Regulatory agencies in the United States and European Union approved injectable forms in 2017 and 2018 respectively. An oral tablet formulation received approval later, using a carrier molecule to enhance absorption across the gastric epithelium. Labeling differs by jurisdiction and by indication.
Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 or 280 nanometres is widely used to assess purity and to resolve related impurities. Liquid chromatography coupled to mass spectrometry confirms identity through the protonated molecular ion and fragment ions formed in tandem experiments. Capillary electrophoresis and peptide mapping after enzymatic digestion supply complementary information on charge variants and modification sites. Circular dichroism and nuclear magnetic resonance can report on secondary structure in solution. Absolute quantification usually depends on an external standard, and reported purity depends on the detection wavelength and integration parameters chosen.
Lyophilised material appears as a white to off-white cake or powder that is hygroscopic, and containers are usually equilibrated to room temperature before opening to limit condensation. Dissolution is performed in water, phosphate-buffered saline, or a mildly alkaline buffer, since solubility rises above neutral pH. Gentle inversion or low-speed mixing is preferred, because vigorous vortexing can promote surface denaturation and aggregation. Complete dissolution may require several minutes, and brief sonication is sometimes applied. Passing the solution through a 0.22 micrometre membrane removes particulates but does not by itself sterilise the liquid.

Stability studies examine how temperature, pH, and moisture influence degradation rates over time. In aqueous solution, hydrolysis and deamidation accelerate as pH moves away from mildly acidic conditions. Light exposure and residual metal ions can also trigger oxidation of susceptible residues. Accelerated aging at elevated temperature is used to estimate shelf life, though extrapolation to room temperature carries uncertainty because individual degradation pathways do not always scale predictably.
Lyophilized semaglutide powder is typically held at minus twenty degrees Celsius for long-term storage. At that temperature, solid-state degradation reactions proceed slowly and the peptide remains intact for extended periods. Repeated freeze-thaw cycles are best avoided because they promote aggregation and can shift the proportion of monomeric peptide present. Working aliquots are often prepared so that each portion is thawed only once, and desiccant is placed inside the vial to limit moisture uptake.
Semaglutide dissolves readily in water and in aqueous buffers near neutral pH. Solubility decreases near the isoelectric point, where net charge is minimal. Common laboratory solvents include phosphate-buffered saline and dilute ammonium bicarbonate. Strongly acidic or basic conditions may accelerate hydrolysis. Working concentrations are usually prepared by diluting a concentrated stock. Vial surfaces can adsorb small amounts of peptide at low concentrations, so carrier proteins or low-binding tubes are sometimes used.
Reverse-phase high-performance liquid chromatography is the standard method for purity assessment, separating the peptide from truncated or oxidized variants. Mass spectrometry confirms molecular mass and detects modifications, while ultraviolet absorbance near 280 nanometers supports concentration measurement through tryptophan and tyrosine residues. Circular dichroism can indicate secondary structure, though the peptide is largely helical in solution, and ion-exchange chromatography resolves charge variants. Purity values above 95 percent are typical for research-grade material. Stability studies track degradation over time under defined conditions.
Lyophilized semaglutide is typically stored at temperatures between minus 20 and minus 80 degrees Celsius for long-term preservation. Short-term storage at 2 to 8 degrees Celsius is common for working aliquots. Repeated freeze-thaw cycles can degrade the peptide and are usually avoided. The molecule is hygroscopic in its solid form, so containers should remain sealed with desiccant. Solutions are less stable than powders and are generally prepared fresh. Light exposure is limited because aromatic residues can undergo photo-oxidation.
Activation of the GLP-1 receptor couples to Gs signalling and raises intracellular cyclic AMP in pancreatic beta cells. The resulting insulin release depends on prevailing glucose concentrations, so the effect is greater when glucose is elevated. Receptor engagement also suppresses glucagon secretion and slows gastric emptying, which flattens post-meal glucose excursions. In the central nervous system, signalling in hypothalamic and brainstem regions is associated with reduced appetite and lower energy intake. Studies continue to examine effects on cardiac, renal and hepatic endpoints; whether those benefits are independent of weight change remains an open question.
Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.
=== Continued employment by the Astros === As part of a personal services contract Bagwell signed with the Astros, he served as a coach in spring training of 2007. On July 11, 2010, the Astros hired Bagwell to be their hitting coach, replacing Sean Berry. At the time of the switch, the Astros had the second-worst average (.237) in the majors and the worst OBP (.295). At the end of the season, the team marginally improved, but was still last in the league in OBP (.303) and SLG (.362) and Bagwell announced he would not return as hitting coach. After a five-year gap in contact with the Astros organization, Bagwell accepted a formal invitation from manager A. J. Hinch to be a guest instructor in spring training of 2015. He is currently a special advisor to team owner Jim Crane.

==== Bile ==== Bile produced by the liver is made up of water (97%), bile salts, mucus and pigments, 1% fats and inorganic salts. Bilirubin is its major pigment. Bile acts partly as a surfactant which lowers the surface tension between either two liquids or a solid and a liquid and helps to emulsify the fats in the chyme. Food fat is dispersed by the action of bile into smaller units called micelles. The breaking down into micelles creates a much larger surface area for the pancreatic enzyme, lipase to work on. Lipase digests the triglycerides which are broken down into two fatty acids and a monoglyceride. These are then absorbed by villi on the intestinal wall. If fats are not absorbed in this way in the small intestine problems can arise later in the large intestine which is not equipped to absorb fats. Bile also helps in the absorption of vitamin K from the diet. Bile is collected and delivered through the common hepatic duct. This duct joins with the cystic duct to connect in a common bile duct with the gallbladder. Bile is stored in the gallbladder for release when food is discharged into the duodenum and also after a few hours.

=== 31 May === One person was killed by Russian shelling in Vovchansk Raion in Kharkiv Oblast, while three people, including two children, were injured by Russian shelling in Kherson Oblast. One person was also killed by Russian shelling in Donetsk Oblast. Officials in the Russian-installed Luhansk People's Republic said that five people were killed and 19 others were injured after Ukrainian artillery hit a poultry farm. The State Border Guard Service of Ukraine reported that Russian troops had blown up a road that links Ukraine's Chernihiv Oblast with Russia's Bryansk Oblast, saying that the incident occurred at the tripoint between Ukraine, Russia and Belarus. Ukraine claimed that Russian-installed officials in occupied Luhansk were threatening to withhold the wages of workers who refused to open accounts at Russian banks. The Russian defence ministry claimed its forces had pushed out Ukrainian forces from positions around Krasnohorivka and Yasynuvata in Donetsk Oblast. A fire broke out at the Afipsky oil refinery in Russia's Krasnodar Krai. Veniamin Kondratyev, Governor of the region, blamed the fire on a Ukrainian drone strike. The fire was put out with minimal damage and no casualties. Another drone was reported to have crashed into the Ilsky refinery. The governor of Belgorod Oblast announced the beginning of evacuations of children from areas affected by Ukrainian shelling, saying that 300 children would be taken to Voronezh, about 250 km (160 mi) away, while another 1,000 children would be evacuated to other regions over the coming days.
=== Ligament and tendon repair === Autologous stem cell-based treatments for ligament injury, tendon injury, osteoarthritis, osteochondrosis, and sub-chondral bone cysts have been commercially available to practicing veterinarians to treat horses since 2003 in the United States and since 2006 in the United Kingdom. Autologous stem cell based treatments for tendon injury, ligament injury, and osteoarthritis in dogs have been available to veterinarians in the United States since 2005. Over 3000 privately owned horses and dogs have been treated with autologous adipose-derived stem cells. The efficacy of these treatments has been shown in double-blind clinical trials for dogs with osteoarthritis of the hip and elbow and horses with tendon damage. Race horses are especially prone to injuries of the tendon and ligaments. Conventional therapies are very unsuccessful in returning the horse to full functioning potential. Natural healing, guided by the conventional treatments, leads to the formation of fibrous scar tissue that reduces flexibility and full joint movement. Traditional treatments prevented a large number of horses from returning to full activity and also have a high incidence of re-injury due to the stiff nature of the scarred tendon. Introduction of both bone marrow and adipose derived stem cells, along with natural mechanical stimulus promoted the regeneration of tendon tissue. The natural movement promoted the alignment of the new fibers and tendocytes with the natural alignment found in uninjured tendons.
In the 1954 version of Casino Royale, Bond was an American agent working with the British to destroy a ruthless Soviet agent in France, but became more widely known as Agent 007, James Bond, of Her Majesty's Secret Service, who was played by Sean Connery until 1971 and by several actors since. Although Bond films often used the Cold War as a backdrop, the Soviet Union itself was almost never Bond's enemy, that role being more often left to fictional and apolitical criminal organisations (like the infamous SPECTRE). However, Red China was in league with Bond's enemies in the films Goldfinger, You Only Live Twice and The Man With the Golden Gun, while some later movies (Octopussy, The Living Daylights) featured a rogue Soviet general as the enemy. TASS Upolnomochen Zayavit... (TASS is Authorized to Announce ... ) – a Soviet TV series based on Julian Semenov's novel. The plot of the movie is set around fictional African country Nagonia, where CIA agents are preparing a military coup, while KGB agent Slavin is trying to prevent it. Slavin succeeds by blackmailing the corrupt American spy John Glebe. The Falcon and the Snowman is a 1985 film directed by John Schlesinger about two young American men, Christopher Boyce and Daulton Lee, who sold United States security secrets to the Soviet Union. The film is based upon the 1979 book The Falcon and the Snowman: A True Story of Friendship and Espionage by Robert Lindsey.

== Mechanical properties == To ensure durability of PCs, mechanical properties are important to study. Elaborate efforts have been made for studying compressive brittleness of porous carbon materials. In 1999, Iizuka, et al. studied the mechanical properties of wood ceramics, a type of porous carbon material. Stable medium-density fiber was used as the base material of wood ceramics and phenol resin was impregnated into the board. Starting at 300 °C, Young's modulus and the compressive strength first decreased with increasing temperature, but at 500 °C the strength increases sharply until it reaches 800 °C and plateaus. The effects of temperature were due to microstructural changes in the resin during carbonization. Effects of impregnates phenol resin at 800 °C were also investigated. Results showed that Young's modulus increased with phenol resin impregnation (Figure 1). The maximum Young's modulus was 5 MPa and the maximum compressive strength was 80 MPa. Wall-bending mechanical test were also performed and it was found that cell wall is breakage was correlated to relative density on compressive strength and Young's modulus. Another type of compressive porous carbon consisting of cellulose and graphene aerogels was studied by Mi, et al. Modified cellulose/graphene aerogels (MCGA) was synthesized via bidirectional freeze drying and grafting of long carbon chains through chemical vapor deposition (Figure 2). The final product had a bulk density of 5.9 mg/cm3 and surface area of 47.3 m2/g with flexible cellulose nanofibril and stiff graphene components.

It is a modified version of the natural hormone, with three amino acid changes and a fatty acid side chain added. These edits extend its half-life from minutes to about one week. The core receptor activity is retained.
Both deliver the same active peptide and act on the same receptor. The tablet includes an absorption enhancer because peptides are poorly taken up intact from the gut. Bioavailability of the oral route is substantially lower, so the two are not dose-equivalent.
No, it is entirely synthetic and does not occur in nature. Native GLP-1 is produced in the gut and pancreas, but the analog is manufactured by chemical synthesis or recombinant methods. Traces of the analog are not expected in people who never received it.
Filtering removes particulate matter that can block columns or scatter light. A 0.22 micrometre membrane is typical, and the filter material should be checked for peptide adsorption.
