This is a working overview of deamidation, written for readers who want more than a one-paragraph summary but less than a textbook.
This page was last updated on 2025-08-29 and is reviewed periodically as new material appears.
Stability studies examine how temperature, pH, and moisture influence degradation rates over time. In aqueous solution, hydrolysis and deamidation accelerate as pH moves away from mildly acidic conditions. Light exposure and residual metal ions can also trigger oxidation of susceptible residues. Accelerated aging at elevated temperature is used to estimate shelf life, though extrapolation to room temperature carries uncertainty because individual degradation pathways do not always scale predictably.
Lyophilized semaglutide powder is typically held at minus twenty degrees Celsius for long-term storage. At that temperature, solid-state degradation reactions proceed slowly and the peptide remains intact for extended periods. Repeated freeze-thaw cycles are best avoided because they promote aggregation and can shift the proportion of monomeric peptide present. Working aliquots are often prepared so that each portion is thawed only once, and desiccant is placed inside the vial to limit moisture uptake.
Identity and purity are usually assessed by reverse-phase high-performance liquid chromatography coupled to mass spectrometry. Retention time and observed mass are compared against a reference standard run under identical conditions. Impurity profiles reveal deamidation products, oxidized methionine variants, and truncated fragments that arise during synthesis or storage. Peptide mapping through enzymatic digestion confirms the primary sequence, while amino acid analysis offers an independent check on overall composition.
Handling practices center on minimizing contamination and adsorption. Lyophilized peptide tends to accumulate static charge, so weighing is done with antistatic measures and calibrated balances. Reconstitution with appropriate solvent should be gentle, avoiding vigorous vortexing that generates foam and shear. Solutions are typically aliquoted before freezing to reduce repeated temperature cycling. Personal protective equipment and a fume hood are standard for powder handling.
Reconstituted solutions are less stable than the dry powder, and stability depends on concentration, pH, buffer composition, and container material. Low-protein-binding tubes reduce loss of peptide to plastic surfaces. Some researchers add a carrier protein to limit adsorption at low concentrations. The exact shelf life of a given solution is best determined empirically through a stability study rather than assumed from general guidance, because published data cover only a limited set of conditions.

Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released after nutrient intake. The molecule contains 31 amino acid residues and differs from the native sequence at several positions. A non-natural residue at position eight resists the enzyme that normally truncates the hormone, while a lysine-linked fatty diacid side chain promotes binding to serum albumin. These two modifications extend the circulating half-life from minutes to roughly one week. The peptide is produced by solid-phase synthesis followed by selective acylation, and its identity and purity are confirmed by spectrometric and chromatographic techniques.
The primary target is the GLP-1 receptor, a class B G protein-coupled receptor expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises intracellular cyclic AMP, which potentiates glucose-dependent insulin secretion and lowers glucagon release when blood glucose is elevated. Signalling in the hypothalamus and brainstem is associated with reduced appetite and slower gastric emptying. Because the insulinotropic effect depends on prevailing glucose levels, the hypoglycaemic risk of the peptide alone is described as low in most study settings. The relative contribution of peripheral and central actions remains an active research question.
As a peptide, semaglutide is sensitive to conditions that break amide bonds or modify side chains. Deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and non-covalent aggregation are the main degradation routes described in published stability work. Rate depends strongly on pH, buffer species, ionic strength, temperature and exposure to light. Formulators therefore choose a defined solution pH and often add excipients such as phosphate buffer, propylene glycol and phenol, each of which plays a separate role in pH control, tonicity or preservation.
Storage guidance for the finished injectable product distinguishes the unused state from the in-use state. Before first use, pens are kept refrigerated between 2 and 8 degrees Celsius, protected from light, and never frozen, since freezing can disrupt the peptide or the device. After first use, label instructions in several markets permit storage at room temperature up to about 30 degrees Celsius for a limited number of days. Solid research-grade material is normally held at or below minus 20 degrees Celsius, often with desiccant, and allowed to equilibrate before opening.
Quantification and purity assessment rely on separation methods coupled to optical or mass detection. Reversed-phase high-performance liquid chromatography resolves the intact peptide from related impurities and is the standard assay technique. Size-exclusion chromatography measures aggregates, while ion-exchange chromatography separates charge variants produced by deamidation. Mass spectrometry confirms identity and detects mass shifts of a few daltons. In biological matrices, liquid chromatography with tandem mass spectrometry is often used because immunoassays can cross-react with endogenous GLP-1 or with circulating fragments.
== Design considerations == Virtual reality games are designed to enhance immersion—the perception that one is actually in the virtual world—and presence—the psychological effect that they are actually interacting with the virtual world outside of their physical bodies—concepts which cannot readily be done with traditional "flat screen" games played on a computer monitor or television. A limiting factor for VR games until the 2010s was the overall system latency between a player's actions and the feedback they saw on the headset. For VR to be felt as an immersive experience, the latency needs to be as small as possible so that the player sees feedback in real-time soon following their actions. Technology bottlenecks had been from two major components of VR systems. One area was the rendering speed of computer hardware to update the 3D displays at a fast-enough frame rate. Frame rates of 20 Hz or less appear to most users as a series of separate images rather than continuous video stream, which breaks immersion. In the late 1990s, this computational power could only reasonably be delivered by high-performance workstations such as those from Sun Microsystems and Silicon Graphics. Since then, improvements in graphics processor technology and game engines with optimized rendering systems give consumer-grade hardware the capacity to perform high-speed real-time 3D rendering at 60 Hz or greater at resolutions appropriate for VR applications. The second bottleneck is the processing time to convert tracking sensor information into feedback that is incorporated into the game.
=== Plant breeding === Selective breeding of cannabis plants has expanded and diversified as commercial and therapeutic markets develop. Some growers in the US succeeded in lowering the proportion of CBD-to-THC to accommodate customers who preferred varietals that were more mind-altering due to the higher THC and lower CBD content. In the US, hemp is classified by the federal government as cannabis containing no more than 0.3% THC by dry weight. This classification was established in the 2018 Farm Bill and was refined to include hemp-sourced extracts, cannabinoids, and derivatives in the definition of hemp.
== Derivatives == Coicenals A-C share the common carbon skeleton of 10-(sec-butyl)-6-hydroxy-1,7,9-trimethyl-1,6,7,8,9,9a-hexahydro-1,4-methanobenzo[d]oxepin-2(4H)-ylidene)acetaldehyde. The molecular formulas of each derivative were determined via high-resolution electrospray ionization mass spectrometry. The R group of Coicenal A is a hydrogen. Coicenal B contains a succinate group for its R group. The R group of Coicenal C is a methyl succinate group. Coicenal D contains a different carbon skeleton than A-C and a succinate group for its R group.
The 5 body regions are right upper, left upper, right lower, left lower, and axial. The Widespread Pain Index (WPI) was introduced by the American College of Rheumatology in 2010. It measures the number of body regions experiencing pain, out of a total of 19: left and right shoulder girdle, upper arm, lower arm, hip/buttock/trochanter, upper leg, lower leg, and jaw; plus the chest, abdomen, neck, upper back and lower back. The Symptom Severity Scale (SSS) assesses the severity of six symptoms; fatigue (score 0-3, for no problem, mild, moderate and severe), trouble thinking or remembering (0-3), waking up tired (unrefreshed) (0-3), pain or cramps in lower abdomen (0-1), depression (0-1) and headache (0-1). As of 2022, among diagnosis methods in the US, the ACR 2016 criteria have been judged as the best FM diagnosis criteria available. The UK RCP also recommends these criteria for FM diagnosis. A similar diagnostic approach is taken in Germany. In 2019, the American Pain Society in collaboration with the U.S. Food and Drug Administration developed a new diagnostic system using two dimensions. The first dimension included core diagnostic criteria, and the second included common features. The core diagnostic criteria are:

PET scanning with the radiotracer [18F]fluorodeoxyglucose (FDG) is widely used in clinical oncology. FDG is a glucose analog that is taken up by glucose-using cells and phosphorylated by hexokinase (whose mitochondrial form is significantly elevated in cancer). Metabolic trapping of the radioactive glucose molecule allows the PET scan to be utilized. The concentrations of imaged FDG tracer indicate tissue metabolic activity as it corresponds to the regional glucose uptake. FDG is used to explore the possibility of cancer spreading to other body sites (cancer metastasis). These FDG PET scans for detecting cancer metastasis are the most common in standard medical care (representing 90% of current scans). The same tracer may also be used for the diagnosis of types of dementia. Less often, other radioactive tracers, usually but not always labelled with fluorine-18 (18F), are used to image the tissue concentration of different kinds of molecules of interest inside the body. Because the hydroxy group that is replaced by fluorine-18 to generate FDG is required for the next step in glucose metabolism in all cells, no further reactions occur in FDG. Furthermore, most tissues (with the notable exception of liver and kidneys) cannot remove the phosphate added by hexokinase. This means that FDG will remain trapped in any cell that takes it up until it decays, since phosphorylated sugars, due to their ionic charge, cannot exit from the cell.
Interstitial lung disease (ILD), or diffuse parenchymal lung disease (DPLD), is a group of respiratory diseases affecting the interstitium (the tissue) and space around the alveoli (air sacs) of the lungs. It concerns alveolar epithelium, pulmonary capillary endothelium, basement membrane, and perivascular and perilymphatic tissues. It may occur when an injury to the lungs triggers an abnormal healing response. Ordinarily, the body generates just the right amount of tissue to repair damage, but in interstitial lung disease, the repair process is disrupted, and the tissue around the air sacs (alveoli) becomes scarred and thickened. This makes it more difficult for oxygen to pass into the bloodstream. The disease presents itself with the following symptoms: shortness of breath, nonproductive coughing, fatigue, and weight loss, which tend to develop slowly, over several months. While many forms are progressive and serious, some types of ILD remain mild or stable for extended periods, especially with early detection and appropriate treatment. The average rate of survival for someone with this disease is between three and five years. The term ILD is used to distinguish these diseases from obstructive airways diseases. There are specific types in children, known as children's interstitial lung diseases. The acronym ChILD is sometimes used for this group of diseases. In children, the pathophysiology involves a genetic component, exposure-related injury, autoimmune dysregulation, or all of the components.
As zolpidem is associated with drug tolerance and substance dependence, its prescription guidelines are only for severe insomnia and short periods of use at the lowest effective dose. Tolerance to the effects of zolpidem can develop in some people in just a few weeks. Abrupt withdrawal may cause delirium, seizures, or other adverse effects, especially if used for prolonged periods and at high doses. When drug tolerance and physical dependence to zolpidem develop, treatment usually entails a gradual dose reduction over a period of months to minimize withdrawal symptoms, which can resemble those seen during benzodiazepine withdrawal. Failing that, an alternative method may be necessary for some people, such as a switch to a benzodiazepine equivalent dose of a longer-acting benzodiazepine drug, as for diazepam or chlordiazepoxide, followed by a gradual reduction in dose of the long-acting benzodiazepine. In people who are difficult to treat, an inpatient flumazenil administration allows for rapid competitive binding of flumazenil to GABAA–receptor as an antagonist, thus stopping (and effectively detoxifying) zolpidem from being able to bind as an agonist on GABAA–receptor; slowly drug dependence or addiction to zolpidem will wane. Alcoholics or recovering alcoholics may be at increased risk of physical dependency or abuse of zolpidem. It is not typically prescribed to people with a history of alcoholism, recreational drug use, physical dependency, or psychological dependency on sedative-hypnotic drugs.

The 1860 civil conflict in Mount Lebanon and Damascus (also called the 1860 Syrian Civil War) was a civil conflict in Mount Lebanon during Ottoman rule in 1860–1861 fought mainly between the local Druze and Christians. Following decisive Druze victories and massacres against the Christians, the conflict spilled over into other parts of Ottoman Syria, particularly Damascus, where thousands of Christian residents (10,0000) were killed by Muslim and Druze militiamen. The fighting precipitated a French-led international military intervention. Bitter conflicts between Christians and Druzes, which had been simmering under Ibrahim Pasha's rule (mostly centred on the firmans of 1839 and, more decisively, of 1856, which equalised the status of Muslim and non-Muslim subjects, the former resenting their implied loss of superiority) resurfaced under the new emir (Bashir Shihab III). The sultan deposed Bashir III on 13 January 1842 and appointed Omar Pasha as governor of Mount Lebanon. Representatives of the European powers proposed to the sultan that Mount Lebanon be partitioned into Christian and Druze sections. On 7 December 1842, the sultan adopted the proposal and asked the governor of Damascus to divide the region into two districts: a northern district under a Christian deputy governor and a southern district under a Druze deputy governor. The arrangement came to be known as the "Double Qaimaqamate". Both officials were to be responsible to the governor of Sidon, who resided in Beirut. The Beirut-Damascus highway was the dividing line between the two districts.
=== Electronic Submissions Gateway (ESG) === To standardize, automate and streamline the flow of regulatory data, FDA introduced an Electronic Submissions Gateway (ESG) in 2006. This gateway allows reporting organizations to send regulatory submissions to different centers over the internet, packaged in a center-specific format and enveloped as a GNU-compatible .tar.gz file, through either a FDA-specific WebTrader application or via a more generic B2B communication protocol called AS2 (Applicability Statement 2).
=== Modern research === In August 2018, scientists announced the transformation of gaseous deuterium into a liquid metallic form. This may help researchers better understand gas giant planets, such as Jupiter, Saturn and some exoplanets, since such planets are thought to contain a lot of liquid metallic hydrogen, which may be responsible for their observed powerful magnetic fields.

Manufacturers commonly state multi-year stability when the powder is kept dry and frozen. Actual shelf life depends on residual moisture, vial sealing, and storage temperature. A re-test by chromatography is the only way to confirm remaining purity.
Deamidated and oxidized forms are the most frequently reported impurities. Truncated peptide fragments from incomplete synthesis also appear at low levels. Their relative abundance grows with time, heat exposure, and unfavorable pH.
Retention time matching on a validated column provides strong circumstantial evidence. It does not distinguish compounds with similar hydrophobicity. Mass measurement or peptide mapping is needed for unambiguous identification.
Sealed, protected from light, and refrigerated at two to eight degrees Celsius for most research material. Desiccated storage limits moisture uptake. Allow the vial to reach room temperature before opening to prevent condensation.
