Peptide degradation follows several routes. Hydrolysis cleaves the backbone at susceptible residues, oxidation targets methionine and tryptophan side chains, and aggregation produces higher-molecular-weight species that are difficult to reverse. Light exposure accelerates oxidation, which is why amber glass or opaque secondary packaging is common. Repeated freeze-thaw cycles promote aggregation and are best avoided. Stability-indicating methods detect these changes before they become visible.
Quality control for research material typically involves reversed-phase HPLC for purity and identity, mass spectrometry for molecular weight confirmation, and Karl Fischer titration for residual water content. Peptide content is often reported as the mass of actual peptide rather than total powder mass, since counterions and water contribute to the latter. A certificate of analysis should list the method used for each specification. Limits and acceptance criteria vary by supplier and by intended application.
Handling practices center on minimizing contamination and adsorption. Lyophilized peptide tends to accumulate static charge, so weighing is done with antistatic measures and calibrated balances. Reconstitution with appropriate solvent should be gentle, avoiding vigorous vortexing that generates foam and shear. Solutions are typically aliquoted before freezing to reduce repeated temperature cycling. Personal protective equipment and a fume hood are standard for powder handling.
Three structural changes define the molecule. At position 8 an alpha-aminoisobutyric acid residue replaces alanine, which blocks dipeptidyl peptidase-4 cleavage. At position 34 arginine replaces lysine, and at position 26 a lysine carries a C18 fatty diacid attached through a short linker. The fatty chain binds serum albumin, and this albumin association reduces renal filtration and enzymatic attack. The unchanged backbone retains the receptor contacts that produce signalling. The free base has the formula C187H291N45O59 and a molecular weight near 4114 daltons.
Receptor activation follows the canonical Gs pathway: binding increases intracellular cyclic AMP, which promotes protein kinase A activity. In pancreatic beta cells this amplifies glucose-dependent insulin release, so secretion rises when blood glucose is high and changes little when it is low. The same signalling suppresses glucagon release from alpha cells and slows gastric emptying. Receptors in the hypothalamus and brainstem are thought to contribute to reduced appetite and lower energy intake. Which of these effects dominates clinical outcomes remains an area of active study.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released by intestinal L cells after food intake. The natural hormone acts on pancreatic and central receptors but is degraded within minutes by dipeptidyl peptidase-4 and other peptidases. Semaglutide belongs to the class of long-acting GLP-1 receptor agonists, a group distinguished by structural changes that slow breakdown and extend circulation time. Its development followed earlier short-acting analogues and reflects a general strategy in peptide drug design: preserve receptor activity while blocking proteolytic clearance.

Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 or 280 nanometres is widely used to assess purity and to resolve related impurities. Liquid chromatography coupled to mass spectrometry confirms identity through the protonated molecular ion and fragment ions formed in tandem experiments. Capillary electrophoresis and peptide mapping after enzymatic digestion supply complementary information on charge variants and modification sites. Circular dichroism and nuclear magnetic resonance can report on secondary structure in solution. Absolute quantification usually depends on an external standard, and reported purity depends on the detection wavelength and integration parameters chosen.
Lyophilised material appears as a white to off-white cake or powder that is hygroscopic, and containers are usually equilibrated to room temperature before opening to limit condensation. Dissolution is performed in water, phosphate-buffered saline, or a mildly alkaline buffer, since solubility rises above neutral pH. Gentle inversion or low-speed mixing is preferred, because vigorous vortexing can promote surface denaturation and aggregation. Complete dissolution may require several minutes, and brief sonication is sometimes applied. Passing the solution through a 0.22 micrometre membrane removes particulates but does not by itself sterilise the liquid.
Storage guidance for the finished injectable product distinguishes the unused state from the in-use state. Before first use, pens are kept refrigerated between 2 and 8 degrees Celsius, protected from light, and never frozen, since freezing can disrupt the peptide or the device. After first use, label instructions in several markets permit storage at room temperature up to about 30 degrees Celsius for a limited number of days. Solid research-grade material is normally held at or below minus 20 degrees Celsius, often with desiccant, and allowed to equilibrate before opening.
Quantification and purity assessment rely on separation methods coupled to optical or mass detection. Reversed-phase high-performance liquid chromatography resolves the intact peptide from related impurities and is the standard assay technique. Size-exclusion chromatography measures aggregates, while ion-exchange chromatography separates charge variants produced by deamidation. Mass spectrometry confirms identity and detects mass shifts of a few daltons. In biological matrices, liquid chromatography with tandem mass spectrometry is often used because immunoassays can cross-react with endogenous GLP-1 or with circulating fragments.
As a peptide, semaglutide is sensitive to conditions that break amide bonds or modify side chains. Deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and non-covalent aggregation are the main degradation routes described in published stability work. Rate depends strongly on pH, buffer species, ionic strength, temperature and exposure to light. Formulators therefore choose a defined solution pH and often add excipients such as phosphate buffer, propylene glycol and phenol, each of which plays a separate role in pH control, tonicity or preservation.
Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.
In some reactions, protons and hydroxide may directly act as acid and base in term of specific acid and specific base catalysis. But more often groups in substrate and active site act as Brønsted–Lowry acid and base. This is called general acid and general base theory. The easiest way to distinguish between them is to check whether the reaction rate is determined by the concentrations of the general acid and base. If the answer is yes then the reaction is the general type. Since most enzymes have an optimum pH of 6 to 7, the amino acids in the side chain usually have a pKa of 4~10. Candidate include aspartate, glutamate, histidine, cysteine. These acids and bases can stabilise the nucleophile or electrophile formed during the catalysis by providing positive and negative charges.
Photosynthesis is the primary process by which carbon moves from the atmosphere into living things. In photosynthetic pathways 12C is absorbed slightly more easily than 13C, which in turn is more easily absorbed than 14C. The differential uptake of the three carbon isotopes leads to 13C/12C and 14C/12C ratios in plants that differ from the ratios in the atmosphere. This effect is known as isotopic fractionation. To determine the degree of fractionation that takes place in a given plant, the amounts of both 12C and 13C isotopes are measured, and the resulting 13C/12C ratio is then compared to a standard ratio known as PDB. The 13C/12C ratio is used instead of 14C/12C because the former is much easier to measure, and the latter can be easily derived: the depletion of 13C relative to 12C is proportional to the difference in the atomic masses of the two isotopes, so the depletion for 14C is twice the depletion of 13C. The fractionation of 13C, known as δ13C, is calculated as follows:
The inaugural Foundation scholar was nobel laureate Linus Pauling and the Foundation has brought many other eminent research chemists to Australia. The Foundation has thus functioned to increase Australian awareness of state of the art international research, increased international recognition of Australian research, and allowed graduate students to interact with leading chemists based in institutions far from Australia. Many of the students have gone on to post-graduate or post-doctoral positions with a Foundation scholar. Freeman also provided advice to the Australian Government on the problem of access to "big science" facilities. This included making major contributions to the report Small Country - Big Science in his work for the Australian Science and Technology Council. The report emphasised the need for Australian researchers to have access to facilities such as synchrotron X-ray and high intensity neutron sources, and led directly to the formation of the Australian Synchrotron Research Programme (ASRP) to fund access to such facilities. Freeman served as a board member of the ASRP until its functions were subsumed under the newly commissioned Australian Synchrotron in 2008. The Australian expertise developed as a consequence of ASRP-supported research led to the Australian Synchrotron being built a decade sooner than would have otherwise been the case. Freeman retired from his Chair in 1997, and was succeeded by Len Lindoy FAA.
Colombia finished fourth in CONMEBOL with 27 points to reach the 2018 FIFA World Cup. They lost their opening match 2–1 to Japan in Saransk after Carlos Sánchez was sent off for a deliberate handball two minutes and 56 seconds in — the second-fastest dismissal in World Cup history, behind José Batista's for Uruguay in 1986 — then beat Poland 3–0 and Senegal 1–0 to top Group H. In the round of 16 at the Spartak Stadium in Moscow, Yerry Mina equalised in the third minute of stoppage time to force a 1–1 draw with England; England won the shootout 4–3, Mateus Uribe striking the crossbar and Carlos Bacca's attempt being saved.

=== Legal status === In August 2026, Scholar Rock Netherlands withdrew its application for a marketing authorization of Isembyld for the treatment of 5q spinal muscular atrophy. The company stated that the manufacturing site used to make the medicine had not demonstrated compliance with EU GMP requirements within the required time limit. Apitegromab was authorized for medical use in the United States in September 2026. The US Food and Drug Administration (FDA) granted the application for apitegromab fast track, orphan drug, and rare pediatric disease designations. The FDA granted the approval of Isembyld to Scholar Rock.
The LSD molecule has two chiral centers at carbons 5 and 8 of the ergoline ring system and hence there are four possible enantiomeric stereoisomers of LSD. Iso-LSD, also known as d-iso-LSD, (+)-iso-LSD, or (5R-8S)-LSD, is one of four possible stereoisomers. The other isomers are LSD (d-LSD, (+)-LSD, or (5R,8R)-LSD), l-iso-LSD ((–)-iso-LSD or (5S,8R)-iso-LSD), and l-LSD ((–)-LSD or (5S,8S)-LSD). None of them are known to have significant psychoactivity in humans besides LSD. LSD is easily epimerized into iso-LSD with base. Consequently, iso-LSD is a common synthetic contaminant in chemical synthesis of LSD. Iso-LSD can be easily epimerized back into LSD. LSD can degrade into iso-LSD depending on temperature, solvent and pH, among other factors. In clinical studies, up to 30% of LSD administered in capsules has been found to isomerize into iso-LSD. Iso-LSD is said to be a metabolite of LSD in animals and humans. However, according to other sources, iso-LSD not a metabolite of LSD but is instead only a contaminant.
=== Blue Men of Lurgan === The "blue men of Lurgan" were a pair of Lurgan men suffering from what was described as "familial idiopathic methemoglobinemia" who were treated by James Deeny in 1942. Deeny, who would later become the Chief Medical Officer of the Republic of Ireland, prescribed a course of ascorbic acid and sodium bicarbonate. In case one, by the eighth day of treatments, there was a marked change in appearance, and by the twelfth day of treatment, the patient's complexion was normal. In case two, the patient's complexion reached normality over a month-long duration of treatment.
Day of Defeat was followed by Day of Defeat: Source, a remake of the game that runs on Valve's Source engine. The Source remake included significant changes to Day of Defeat's gameplay, new maps, updated graphics, and improved physics. Released on September 26, 2005 to favorable reviews, the game was praised for its gameplay, audio, and graphics. Post-release, the game was supported by Valve with a number of subsequent updates, including versions of the game for OS X and Linux released in 2010 and 2013 respectively.
Extending the work of Vesalius into experiments on still living bodies (of both humans and animals), William Harvey and other natural philosophers investigated the roles of blood, veins and arteries. Harvey's De motu cordis in 1628 was the beginning of the end for Galenic theory, and alongside Santorio Santorio's studies of metabolism, it served as an influential model of quantitative approaches to physiology. In the early 17th century, the micro-world of biology was just beginning to open up. A few lensmakers and natural philosophers had been creating crude microscopes since the late 16th century, and Robert Hooke published the seminal Micrographia based on observations with his own compound microscope in 1665. But it was not until Antonie van Leeuwenhoek's dramatic improvements in lensmaking beginning in the 1670s—ultimately producing up to 200-fold magnification with a single lens—that scholars discovered spermatozoa, bacteria, infusoria and the sheer strangeness and diversity of microscopic life. Similar investigations by Jan Swammerdam led to a new interest in entomology and built the basic techniques of microscopic dissection and staining.

In 2000, Shaw et al. demonstrated that circulating 3α-diol mediates prostate development in tammar wallaby pouch young via conversion to DHT in target tissues. Tammar wallaby pouch young do not show sexually dimorphic circulating levels of T and DHT during prostate development which suggests that another androgenization mechanism was responsible. While 3α-diol's androgen receptor binding affinity is five orders of magnitude lower than DHT (3α-diol is nearly inactive at the androgen receptor), it was known that 3α-diol can be oxidized back to DHT via the action of a number of dehydrogenases. In 2003, Wilson et al. demonstrated that 5α-reductase expression in target tissues enabled a novel pathway from 17OHP to 3α-diol without T or A4 as an intermediate. In 2004, Mahendroo et al. demonstrated that an overlapping novel pathway is operating in mouse testes, generalizing what had been demonstrated in tammar wallaby. The term "backdoor pathway" was coined by Auchus in 2004 and was described as 5α-reduction of 17α-hydroxyprogesterone (17OHP) which is a first step in a pathway that ultimately leads to the production of dihydrotestosterone (DHT). and defined as a route to DHT that: (1) bypasses conventional intermediates androstenedione (A4) and T; (2) involves 5α-reduction of C21 pregnanes to C19 androstanes; and (3) involves the 3α-oxidation of 3α-diol to DHT. The backdoor pathway explains how androgens are produced under certain normal and pathological conditions in humans when the classical androgen pathway cannot fully explain the observed consequences.
== History == B&K Corporation Limited was founded in 2012 in Qingdao, China, and commenced research and development of PDGF drugs in 2013. The company subsequently established a research and development centre in Beijing and a laboratory in Hong Kong. Between 2021 and 2023, the company completed three financing rounds, raising a total of approximately RMB405 million. Investors included Qingdao High-Tech and CDH Investment. Following the Series B round in 2023, the company's post-money valuation reached RMB3.3 billion. In July 2026, the company officially changed its Chinese name from "華芢生物" to "青城新藥".
The Revolt, written by Menachem Begin before he became Prime Minister, tells the story of the Lehi (referred to by their translated name's acronym, F.F.I. "Freedom Fighters of Israel" in early English editions) and Begin's own Irgun fighters during the Jewish insurgency in Palestine. The Revolt influenced groups from a wide variety of ideologies. Some of these groups began to use suicide attacks in the 1990s. The book was allegedly found Osama bin Laden's al-Qaeda organization. Begin praised his militants' willingness to die, "These two wonderful young men greeted the sentence with the singing of Hatikvah". The "two wonderful young men" were the two who blew themselves up in 1947, he described their deaths, "even before they put on their red uniforms. They too held their heads high when they faced their judges… But they did not reach the gallows. They too sang on the threshold of death — a song of faith in God: 'Lord of the world who reigned before creation'. But their song ended with a great explosion which shattered the silence of the prison in occupied Jerusalem". In 1977 the author became prime minister, after previously being in opposition. Historical militants featured prominently in his political speeches. He praised the actions of his militants during the insurgency in Palestine in the 1940s, including the leader of the cell who bombed the King David Hotel (killing 91 people and 1 terrorist). His favourites were, again, the two young militants who blew themselves up in Jerusalem prison in 1947.
== Production == Royal jelly is secreted from the glands in the heads of worker bees and is fed to all bee larvae, whether they are destined to become drones (males), workers (sterile females), or queens (fertile females). After three days, the drone and worker larvae are no longer fed with royal jelly, but queen larvae continue to be fed this special substance throughout their development.
=== Etymology === The word diabetes ( or ) comes from Latin diabētēs, which in turn comes from Ancient Greek διαβήτης (diabētēs), which literally means "a passer through; a siphon". Ancient Greek physician Aretaeus of Cappadocia (fl. 2nd century CE) used that word, with the intended meaning "excessive discharge of urine", as the name for the disease. Ultimately, the word comes from Greek διαβαίνειν (diabainein), meaning "to pass through", which is composed of δια- (dia-), meaning "through" and βαίνειν (bainein), meaning "to go". The word "diabetes" is first recorded in English, in the form diabete, in a medical text written around 1425. The word mellitus ( or ) comes from the classical Latin word mellītus, meaning "mellite" (i.e. sweetened with honey; honey-sweet). The Latin word comes from mell-, which comes from mel, meaning "honey"; sweetness; pleasant thing, and the suffix -ītus, whose meaning is the same as that of the English suffix "-ite". It was Thomas Willis who in 1675 added "mellitus" to the word "diabetes" as a designation for the disease, when he noticed the urine of a person with diabetes had a sweet taste (glycosuria). This sweet taste had been noticed in urine by the ancient Greeks, Chinese, Egyptians, and Indians.

Sealed, protected from light, and refrigerated at two to eight degrees Celsius for most research material. Desiccated storage limits moisture uptake. Allow the vial to reach room temperature before opening to prevent condensation.
Each cycle exposes the peptide to ice-liquid interfaces where unfolding and aggregation can occur. Aggregates may not redissolve and can alter measured activity. Aliquoting before the first freeze reduces the number of cycles any single portion experiences.
It usually reports purity by HPLC, identity by mass spectrometry, peptide content, appearance, and residual moisture or counterion content. The analytical method behind each value should be stated. Acceptance criteria are set by the supplier or the buyer's specification.
Native GLP-1 is a short-lived peptide cleared within one to two minutes by dipeptidyl peptidase-4 and related enzymes. Semaglutide keeps the receptor-binding backbone but adds substitutions and a lipid chain. These changes block the main cleavage site and allow reversible albumin binding, extending the half-life to roughly 165 hours.
