Quantification and purity assessment rely on separation methods coupled to optical or mass detection. Reversed-phase high-performance liquid chromatography resolves the intact peptide from related impurities and is the standard assay technique. Size-exclusion chromatography measures aggregates, while ion-exchange chromatography separates charge variants produced by deamidation. Mass spectrometry confirms identity and detects mass shifts of a few daltons. In biological matrices, liquid chromatography with tandem mass spectrometry is often used because immunoassays can cross-react with endogenous GLP-1 or with circulating fragments.
As a peptide, semaglutide is sensitive to conditions that break amide bonds or modify side chains. Deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and non-covalent aggregation are the main degradation routes described in published stability work. Rate depends strongly on pH, buffer species, ionic strength, temperature and exposure to light. Formulators therefore choose a defined solution pH and often add excipients such as phosphate buffer, propylene glycol and phenol, each of which plays a separate role in pH control, tonicity or preservation.

Stability studies examine how temperature, pH, and moisture influence degradation rates over time. In aqueous solution, hydrolysis and deamidation accelerate as pH moves away from mildly acidic conditions. Light exposure and residual metal ions can also trigger oxidation of susceptible residues. Accelerated aging at elevated temperature is used to estimate shelf life, though extrapolation to room temperature carries uncertainty because individual degradation pathways do not always scale predictably.
Lyophilized semaglutide powder is typically held at minus twenty degrees Celsius for long-term storage. At that temperature, solid-state degradation reactions proceed slowly and the peptide remains intact for extended periods. Repeated freeze-thaw cycles are best avoided because they promote aggregation and can shift the proportion of monomeric peptide present. Working aliquots are often prepared so that each portion is thawed only once, and desiccant is placed inside the vial to limit moisture uptake.
Identity and purity are usually assessed by reverse-phase high-performance liquid chromatography coupled to mass spectrometry. Retention time and observed mass are compared against a reference standard run under identical conditions. Impurity profiles reveal deamidation products, oxidized methionine variants, and truncated fragments that arise during synthesis or storage. Peptide mapping through enzymatic digestion confirms the primary sequence, while amino acid analysis offers an independent check on overall composition.
Quality control relies on pharmacopoeial monographs where they exist, combined with in-house specifications for identity, purity, water content, and counter-ion composition. Reference standards allow calibration across laboratories, although certified materials for every analogue are not universally obtainable. Batch records, chromatograms, and mass spectra form the documentation trail. Regulatory classification varies by jurisdiction and intended use, and research-grade material differs from pharmaceutical-grade material in testing scope. Analytical uncertainty is often expressed as relative standard deviation across replicate injections.
Lyophilised semaglutide is generally held at -20 °C or below, protected from light and moisture. Reconstituted solutions are typically kept at 2-8 °C and used within a defined window because degradation accumulates over time. Repeated freeze-thaw cycles are discouraged, since each cycle can promote aggregation and reduce monomeric content. Room-temperature stability of the solid has been examined in some studies but remains incompletely characterised for long durations, so cold storage is the conservative default for research material.
Degradation proceeds along several parallel routes. Deamidation of asparagine and glutamine residues generates charged variants that shift retention time in chromatographic analysis. Oxidation targets methionine and can be accelerated by trace metals or dissolved oxygen. Non-covalent aggregation produces dimers, oligomers, and larger species that are difficult to reverse. Isomerisation at aspartate residues is slower but measurable under thermal stress. The distribution among these pathways depends on pH, buffer composition, ionic strength, and the presence of excipients such as sugars or surfactants.
==== Cover-up allegations ==== Fonterra notified the New Zealand government on 5 September and three days later, Prime Minister Helen Clark had Beijing officials alerted directly. News reports began circulating in China on 9 September, and the news was broken internationally by Reuters the following day. The state-controlled media report did not initially identify the company involved. Posts on Chinese social portal Tianya named Sanlu as the culprit, which Sanlu denied. According to a State Council investigation, Sanlu had known about infants made sick by its products as early as December 2007, but no testing was performed until June 2008. It said leading government officials in Shijiazhuang city had failed to report the contamination to provincial and state authorities (until 9 September) in violation of rules on reporting major incidents involving food safety. According to the People's Daily, on 2 August, Sanlu asked the Shijiazhuang city government to assist them in controlling the media's reporting of the recall. According to accounts confirmed by media reports and health officials, the company tried to buy off critics and cover up the contamination. In a memo dated 11 August, Beijing-based public relations agency Teller International advised Sanlu to seek cooperation with major search engines to censor negative information. The agency reportedly had repeatedly contacted key account staff at Baidu and proposed a ¥3 million (US$440,000) budget to screen all negative news.
=== Efficacy === 2,6-Dichloro-1,4-benzoquinone is primarily used in research due to its ability to participate in electron transfer reactions. In photosynthesis, it can act as an artificial electron acceptor in photosystem II. Researchers can measure and analyse electron transport activity in chloroplasts and other photosynthetic systems. The ability to accept electrons in place of natural quinones makes the compound useful for investigating the mechanisms of photosynthetic energy conversion and for evaluating the effects of inhibitors or environmental stress on photosynthetic organisms. In addition, 2,6-DCBQ can inhibit enzymes such as acetylcholinesterase in laboratory experiments and may exhibit antimicrobial or algal toxicity due to its reactive quinone structure. These characteristics are mainly of research interest rather than therapeutic application.
The 25 April 1953 issue of the journal Nature published a series of five articles giving the Watson and Crick double-helix structure DNA and evidence supporting it. The structure was reported in a letter in which they stated, "It has not escaped our notice that the specific pairing we have postulated immediately suggests a possible copying mechanism for the genetic material." This letter was followed by a letter from Franklin and Gosling, which was the first publication of their own X-ray diffraction data and of their original analysis method. Then followed a letter by Wilkins and two of his colleagues, which contained an analysis of in vivo B-DNA X-ray patterns, and which supported the presence in vivo of the Watson and Crick structure. In April 2023, scientists, based on new evidence, concluded that Rosalind Franklin was a contributor and "equal player" in the discovery process of DNA, rather than otherwise, as may have been presented subsequently after the time of the discovery. In 1962, after Franklin's death, Watson, Crick, and Wilkins jointly received the Nobel Prize in Physiology or Medicine. Nobel Prizes are awarded only to living recipients. A debate continues about who should receive credit for the discovery. In an influential presentation in 1957, Crick laid out the central dogma of molecular biology, which foretold the relationship between DNA, RNA, and proteins, and articulated the "adaptor hypothesis". Final confirmation of the replication mechanism that was implied by the double-helical structure followed in 1958 through the Meselson–Stahl experiment.

== Description == Helianthus tuberosus is a herbaceous perennial plant growing to 1.5–3 m (4 ft 11 in – 9 ft 10 in) tall with opposite leaves on the lower part of the stem but alternate towards the top. The leaves have a rough, hairy texture. Larger leaves on the lower stem are broad ovoid-acute and can be up to 30 cm (12 in) long. Leaves higher on the stem are smaller and narrower. The flowers are yellow and produced in capitate flowerheads, which are 5–10 cm (2–4 in) in diameter, with 10–20 ray florets and 60 or more small disc florets. The flowers are briefly fragrant, giving off a light, vanilla-chocolate scent. The tubers are often elongated and uneven, typically 7.5–10 cm (3–3+7⁄8 in) long and 3–5 cm (1–2 in) thick, and vaguely resembling a ginger root in appearance, with a crisp and crunchy texture when raw. They vary in color from pale brown to white, red, or purple.
=== Mothers === For women with PKU, it is important for the health of their children to maintain low Phe levels before and during pregnancy. Though the developing fetus may only be a carrier of the PKU gene, the intrauterine environment can have very high levels of phenylalanine, which can cross the placenta. The child may develop congenital heart disease, growth retardation, microcephaly, and intellectual disability as a result. In most countries, women with PKU who wish to have children are advised to lower their blood Phe levels (typically to between 2 and 6 mg/dL) before they become pregnant, and carefully control their levels throughout the pregnancy. This is achieved by performing regular blood tests and adhering very strictly to a diet, in general, monitored on a day-to-day basis by a specialist metabolic dietitian. In many cases, as the fetus' liver begins to develop and produce PAH normally, the mother's blood Phe levels will drop, requiring an increased intake to remain within the safe range of 2–6 mg/dL. The mother's daily Phe intake may double or even triple by the end of the pregnancy, as a result. When maternal blood Phe levels fall below 2 mg/dL, anecdotal reports indicate that the mothers may experience adverse effects, including headaches, nausea, hair loss, and general malaise. When low phenylalanine levels are maintained for the duration of pregnancy, there are no elevated levels of risk of birth defects compared with a baby born to a non-PKU mother.
The aspartate pathway is an amino acid metabolic pathway present in bacteria and plants that deal with converting aspartate to other amino acids through a series of reactions and intermediates. L-Aspartate-4-semialdehyde serves as one of the first intermediates in the pathway and as an important step of differentiation in the pathway. L-Aspartate-4-semialdehyde is synthesized by the enzyme aspartate semialdehyde dehydrogenase, which catalyzes the following reversible chemical reaction:
=== Charge-exchange chemical ionization === This is also similar to CI and the difference lies in the production of a radical cation with an odd number of electrons. The reagent gas molecules are bombarded with high energy electrons and the product reagent gas ions abstract electrons from the analyte to form radical cations. The common reagent gases used for this technique are toluene, benzene, NO, Xe, Ar and He. Careful control over the selection of reagent gases and the consideration toward the difference between the resonance energy of the reagent gas radical cation and the ionization energy of the analyte can be used to control fragmentation. The reactions for charge-exchange chemical ionization are as follows.

genetic engineering Also genetic modification or genetic manipulation. The direct, deliberate manipulation of an organism's genetic material using any of a variety of biotechnology methods, including the insertion or removal of genes, the transfer of genes within and between species, the mutation of existing sequences, and the construction of novel sequences using artificial gene synthesis. Genetic engineering encompasses a broad set of technologies by which the genetic composition of individual cells, tissues, or entire organisms may be altered for various purposes, commonly in order to study the functions and expression of individual genes, to produce hormones, vaccines, and other drugs, and to create genetically modified organisms for use in research and agriculture.
Caffeine (1,3,7-trimethylxanthine) is the alkaloid most present in green and roasted coffee beans. The content of caffeine is between 1.0% and 2.5% by weight of dry green coffee beans. The content of caffeine does not change during maturation of green coffee beans, but higher caffeine content is found in plants grown at higher altitudes. The solubility of caffeine in water increases with temperature and with the addition of chlorogenic acids, citric acid, or tartaric acid, all of which are present in green coffee beans. For example, 1 g (0.035 oz) of caffeine dissolves in 46 mL (1.6 US fl oz) of water at room temperature, and 5.5 mL (0.19 US fl oz) at 80 °C (176 °F). Trigonelline (N-methyl-nicotinate) is a derivcular-weight peptides and amino acids. Mature coffee contains free amino acids (4.0 mg amino acid/g robusta coffee and up to 4.5 mg amino acid/g arabica coffee). In Coffea arabica, alanine is the amino acid with the highest concentration, i.e., 1.2 mg/g, followed by asparagine of 0.66 mg/g, whereas in C. robusta, alanine is present at a concentration of 0.8 mg/g and asparagine at 0.36 mg/g. Roasted coffee beans do not contain any free amino acids; the amino acids in green coffee beans are degraded under roasting temperature to Maillard products (reaction products between the aldehyde group of sugar and the alpha-amino group of the amino acids). Further, diketopiperazines, e.g. cyclo(proline-proline), cyclo(proline-leucine), and cyclo(proline-isoleucine), are generated from the corresponding amino acids, and are the major source of the bitter taste of roasted coffee.

When the Basilica of Saint-Denis was desecrated by French revolutionaries in 1793, the body of Henry IV was so well preserved it was displayed two days before being thrown in a mass grave, and a new death mask was able to be made. Likewise, Louis XIII was still recognisable thanks to his well preserved moustache. Turenne was so well preserved that looters first thought he was still alive. Maria II of Portugal died in childbirth on 15 November 1853, at the age of 34. Her embalming was conducted the following day and proved to be a complex procedure due to the rapid post-mortem changes in her remains. The Duchess of Ficalho, the queen's lady-in-waiting, provided a primary account of the event in a letter to her brother, the 2nd Count of Lavradio: The sad embalming took place on November 16th, and I was present throughout. The procedure for the Infante and the Queen lasted seven hours. Once that ordeal was over, it was time to dress the body, which was nearly impossible due to the state of dissolution of Her Majesty. Nevertheless, it was done as well as possible, including the placement of the Orders and the Royal Mantle. It was ultimately necessary to close the coffin, for the state of dissolution was beyond description. In 2012, an exhumation of the remains of Amélie of Leuchtenberg (1812–1873), Empress of Brazil, revealed that her body had undergone extensive long-term preservation. Despite being interred for 139 years, the body was found mummified with skin, hair, and internal organs intact.
As of October 2024, Novo was the second most valuable drug company in the world by market capitalization, second only to its competitor Eli Lilly and Company. By July 2025, Novo had fallen to become the fifth most valuable drug company amid rising competition from generic weight loss substitutes. The company named Maziar Mike Doustdar as the new CEO, effective August 7, 2025. In March 2025, the company announced new plans for a direct-to-consumer offering of its Wegovy weight loss drug. The company established a new pharmacy, called NovoCare, which would charge customers $499 per month for access to the drug, less than half the cost of the drug through other pharmaceutical distribution networks. In September 2025, Novo announced that it would reduce its global workforce by 9,000 people, approximately 9% of its total workforce. In October 2025, Novo acquired Akero Therapeutics for $5.2 billion. Akero Therapeutics is a clinical stage company focusing on the treatment of metabolic dysfunction-associated steatohepatitis (MASH).

Ice formation concentrates solutes and can mechanically stress the peptide or damage the delivery device. Thawing afterwards may leave aggregates that are not visible to the eye. Refrigeration keeps the solution above its freezing point while slowing chemical degradation.
