Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, a gut hormone released after meals. Its backbone retains the GLP-1 sequence but incorporates two substitutions that slow enzymatic breakdown by dipeptidyl peptidase-4. A short polyethylene glycol linker and a C18 fatty diacid are attached to the peptide chain, allowing the molecule to bind serum albumin and remain in circulation far longer than the native hormone. The result is a circulating half-life measured in days rather than the minutes typical of endogenous GLP-1.
Receptor activation occurs at GLP-1 receptors distributed across pancreatic islets, the hypothalamus, and the gastrointestinal tract. Binding triggers G protein signaling that raises cyclic AMP and enhances glucose-dependent insulin release. Because the effect depends on prevailing glucose levels, insulin secretion does not rise when blood sugar is already low. Signaling in the brain and gut also influences appetite and gastric emptying, which is why the compound appears in both metabolic and weight-related research literature.
Development began in the early 2010s with the goal of extending GLP-1 activity beyond the brief window achieved by native peptide infusion. The earliest approved formulation was a subcutaneous injection given once weekly. A later oral tablet pairs the peptide with an absorption enhancer, sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, usually shortened to SNAC. That carrier lowers local pH and helps the peptide cross gastric tissue. Both routes deliver the same active molecule.

Certificate of analysis documents from suppliers typically report purity by chromatographic area, water content, and counter-ion identity. Independent verification is advisable because reported values can be generated under differing conditions. Impurity profiles matter for research use, where aggregates, deamidation products, and residual solvents may influence experimental results. Container, lot, and chain-of-custody records support traceability. Analytical results are method-dependent, so comparisons between laboratories require the same procedure and reference standards.
Lyophilized peptide material is typically stored at or below -20 °C, with -80 °C used for longer-term archives. Vials should remain sealed and desiccated because moisture promotes aggregation and hydrolysis. Repeated freeze-thaw cycles are avoided since they can alter peptide conformation and reduce recovery. Once reconstituted, solutions are generally kept at 2-8 °C and used within a defined window. Stability beyond those windows depends on buffer composition and concentration, and exact limits are product-specific rather than universal.
Identity and purity are assessed with reversed-phase high-performance liquid chromatography, which separates the peptide from related impurities by hydrophobicity. Mass spectrometry confirms molecular weight and detects truncation or modification products. Peptide mapping after enzymatic digestion verifies the amino acid sequence. Quantitation is often performed by LC-MS/MS or by immunoassay, and the two approaches can give different values because they measure different things. Method validation parameters such as accuracy, precision, and limit of quantitation are reported alongside results.
Material described as research-grade is not necessarily manufactured to pharmaceutical standards, and purity figures depend on the method used to obtain them. A certificate of analysis states the measured purity, the analytical technique, and the batch identifier, but the underlying data are not always included. Independent testing by a second laboratory is a common way to confirm identity and purity. Uncertainties remain about how storage history affects long-term stability, and about how well results from one laboratory transfer to another. Documentation of handling conditions supports comparison between batches.
Peptides are sensitive to temperature, light, oxygen, and repeated freeze-thaw cycles. Semaglutide in dry form is generally held at refrigerated temperatures, while reconstituted solutions require a defined short-term storage window. Vials should be kept in secondary packaging to limit photodegradation, and exposure to alkaline conditions is avoided because it accelerates chemical degradation. Adsorption to glass and some plastics can reduce the measured concentration of dilute solutions, so low-binding polypropylene containers are preferred for analytical work. Each transfer step introduces a small risk of contamination, and closed handling practices reduce that risk.

A step experiment is often easier to perform than a pulse experiment, but it tends to smooth over some of the details that a pulse response could show. It is easy to numerically integrate an experimental pulse response to obtain a very high-quality estimate of the step response, but the reverse is not the case because any noise in the concentration measurement will be amplified by numeric differentiation.
Upon activation, bAREs ADP-ribosylate any number of eukaryotic proteins; such mechanism is crucial to the instigation of the diseased states associated with ADP-ribosylation. GTP-binding proteins, in particular, are well-established in bAREs pathophysiology. For examples, cholera and heat-labile enterotoxin target the α-subunit of Gs of heterotrimeric GTP-binding proteins. As the α-subunit is ADP-ribosylated, it is permanently in an "active", GTP-bound state; subsequent activation of intracellular cyclic AMP stimulates the release of fluid and ions from intestinal epithelial cells. Furthermore, C. Botulinum C3 ADP-ribosylates GTP-binding proteins Rho and Ras, and Pertussis toxin ADP-ribosylates Gi, Go, and Gt. Diphtheria toxin ADP-ribosylates ribosomal elongation factor EF-2, which attenuates protein synthesis. There are a variety of bacteria which employ bAREs in infection: CARDS toxin of Mycoplasma pneumoniae, cholera toxin of Vibrio cholerae; heat-labile enterotoxin of E. coli; exotoxin A of Pseudomonas aeruginosa; pertussis toxin of B. pertussis; C3 toxin of C. botulinum; and diphtheria toxin of Corynebacterium diphtheriae.
Dinuzulu made a pact with the Boers of his own, promising them land in return for their aid. The Boers were led by Louis Botha. Dinuzulu and the Boers defeated Zibhebhu in 1884. They were granted about half of Zululand individually as farms, and formed the independent Republic of Vryheid. This alarmed the British who wanted to prevent the Boers access to a harbour. The British then annexed Zululand in 1887. Dinuzulu became involved in later conflicts with rivals. In 1906 Dinuzulu was accused of being behind the Bambatha Rebellion. He was arrested and put on trial by the British for "high treason and public violence". In 1909, he was sentenced to ten years' imprisonment on St Helena island. When the Union of South Africa was formed, Louis Botha became its first prime minister, and he arranged for his old ally Dinuzulu to return to South Africa and live in exile on a farm in the Transvaal, where he died in 1913. Dinuzulu's son Solomon kaDinuzulu was never recognised by South African authorities as the Zulu king, only as a local chief, but he was increasingly regarded as king by chiefs, by political intellectuals such as John Langalibalele Dube and by ordinary Zulu people. In 1923, Solomon founded the organisation Inkatha YaKwaZulu to promote his royal claims, which became moribund and then was revived in the 1970s by Mangosuthu Buthelezi, chief minister of the KwaZulu bantustan.

=== Lysosomal damage inhibits mTOR and induces autophagy === Active mTORC1 is positioned on lysosomes. mTOR is inhibited when lysosomal membrane is damaged by various exogenous or endogenous agents, such as invading bacteria, membrane-permeant chemicals yielding osmotically active products (this type of injury can be modeled using membrane-permeant dipeptide precursors that polymerize in lysosomes), amyloid protein aggregates (see above section on Alzheimer's disease) and cytoplasmic organic or inorganic inclusions including urate crystals and crystalline silica. The process of mTOR inactivation following lysosomal/endomembrane damage is mediated by the protein complex termed GALTOR. At the heart of GALTOR is galectin-8, a member of β-galactoside binding superfamily of cytosolic lectins termed galectins, which recognizes lysosomal membrane damage by binding to the exposed glycans on the lumenal side of the delimiting endomembrane. Following membrane damage, galectin-8, which normally associates with mTOR under homeostatic conditions, no longer interacts with mTOR but now instead binds to SLC38A9, RRAGA/RRAGB, and LAMTOR1, inhibiting Ragulator's (LAMTOR1-5 complex) guanine nucleotide exchange function. mTOR is a negative regulator of autophagy in general, best studied during response to starvation, which is a metabolic response. During lysosomal damage however, mTOR inhibition activates autophagy response in its quality control function, leading to the process termed lysophagy that removes damaged lysosomes.
During the opening phase of the Battle of Stalingrad, when the Germans overran the Kuban, the majority of the Cossack population, long before the Germans began their agitation with Krasnov and Shkuro, became involved in Partisan activity. Raids on the German positions from the Caucasus mountains became commonplace. After the German defeat at Stalingrad, the 4th Guards Kuban Cossack Corps, strengthened by tanks and artillery, broke through the German lines and liberated Mineralnye Vody, and Stavropol.
== Characterization == The glutathione peroxidase family consists of 8 known human isoforms. Glutathione peroxidases use glutathione as an electron donor and are active with both hydrogen peroxide and organic hydroperoxide substrates. Gpx1, Gpx2, Gpx3, and Gpx4 have been shown to be selenium-containing enzymes, whereas Gpx6 is a selenoprotein in humans with cysteine-containing homologues in rodents. Amyloid beta, when bound to heme, has been shown to have peroxidase activity. A typical group of peroxidases are the haloperoxidases. This group is able to form reactive halogen species and, as a result, natural organohalogen substances. A majority of peroxidase protein sequences can be found in the PeroxiBase database.
=== Condensed matter and macromolecules === Metastable phases are common in condensed matter and crystallography. This is the case for anatase, a metastable polymorph of titanium dioxide, which despite commonly being the first phase to form in many synthesis processes due to its lower surface energy, is always metastable, with rutile being the most stable phase at all temperatures and pressures. As another example, diamond is a stable phase only at very high pressures, but is a metastable form of carbon at standard temperature and pressure. It can be converted to graphite (plus leftover kinetic energy), but only after overcoming an activation energy – an intervening hill. Martensite is a metastable phase used to control the hardness of most steel. Metastable polymorphs of silica are commonly observed. In some cases, such as in the allotropes of solid boron, acquiring a sample of the stable phase is difficult. The bonds between the building blocks of polymers such as DNA, RNA, and proteins are also metastable. Adenosine triphosphate (ATP) is a highly metastable molecule, colloquially described as being "full of energy" that can be used in many ways in biology. Generally speaking, emulsions/colloidal systems and glasses are metastable. The metastability of silica glass, for example, is characterised by lifetimes on the order of 1098 years (as compared with the lifetime of the universe, which is thought to be around 1.3787×1010 years). Sandpiles are one system which can exhibit metastability if a steep slope or tunnel is present. Sand grains form a pile due to friction.

Native GLP-1 is degraded within minutes by circulating enzymes. The synthetic version carries substitutions at positions that block enzymatic cleavage, plus a fatty acid side chain that promotes albumin binding. These two changes together extend circulation time from minutes to roughly a week.
Albumin binding keeps a large fraction of the compound in a slowly released reservoir within the bloodstream. Plasma levels decline gradually rather than falling sharply after each administration. That profile supports dosing intervals measured in days instead of hours.
The active peptide sequence is the same in both formats. The oral version adds an absorption enhancer that is not present in the injected solution. Differences in excipients and formulation affect uptake rather than the identity of the active molecule.
冻融过程中冰晶形成与局部浓度升高会促使肽链发生界面吸附和聚集。聚集不仅降低有效含量,还会改变可见异物与不溶性微粒的计数结果。将溶液分装为单次使用的小体积等份可减少循环次数。
