Receptor binding triggers G protein signaling that raises intracellular cyclic AMP in pancreatic beta cells. Insulin release follows in a glucose-dependent manner, so secretion increases when blood glucose is elevated and diminishes when it is not. The same signaling suppresses glucagon release from alpha cells and slows gastric emptying, which blunts the post-meal glucose rise. In the brain, receptor activation in regions such as the arcuate nucleus is associated with reduced appetite and lower energy intake. How much each of these effects contributes to overall weight change is not fully settled.
Two structural features account for the prolonged half-life of semaglutide. A modified amino acid at position 8 resists cleavage by dipeptidyl peptidase-4, the enzyme that rapidly degrades native GLP-1. A fatty diacid side chain binds serum albumin, which limits renal clearance and protects the peptide from enzymatic breakdown. These modifications yield a plasma half-life of approximately one week in humans, allowing once-weekly administration. The relationship between plasma concentration and clinical effect varies between individuals, and sources of that variability are still being characterized.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1 (GLP-1), a hormone released from intestinal L-cells after food intake. The compound belongs to the incretin mimetic class and acts at GLP-1 receptors distributed across pancreatic, gastrointestinal, cardiovascular, and central nervous system tissues. Compared with native GLP-1, the molecule carries structural changes that extend its activity from minutes to roughly one week. It is studied for glycemic control in type 2 diabetes and for weight management, and its effects on cardiovascular and other outcomes remain active research areas.
Clinical studies of semaglutide generally measure glycated hemoglobin, fasting plasma glucose, body weight, and composite cardiovascular endpoints. The SUSTAIN program enrolled adults with type 2 diabetes, while the STEP program focused on obesity without diabetes. Administration follows a stepwise escalation schedule designed to limit gastrointestinal effects during the first weeks. Reported outcomes include mean percentage weight change, the proportion of participants reaching defined weight-loss thresholds, and rates of nausea, vomiting, and diarrhea. Long-term data on durability after treatment stops are still limited and remain a topic of ongoing research.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, developed by Novo Nordisk and first approved in 2017 for type 2 diabetes. It belongs to the incretin mimetic class, a group of agents that reproduce the glucose-dependent actions of endogenous GLP-1. The molecule was engineered to resist degradation by dipeptidyl peptidase-4 and to bind serum albumin, extending its half-life from minutes to roughly one week. Approval for chronic weight management followed in 2021, based on large cardiovascular and obesity outcome trials.

Semaglutide is a synthetic peptide analog of glucagon-like peptide-1, a hormone released from intestinal L cells after food intake. The molecule is a 31-amino-acid backbone modified at three positions to resist cleavage by dipeptidyl peptidase-4, the enzyme that degrades native GLP-1 within minutes. A lysine residue at position 26 carries a linker and a C18 fatty diacid, which promotes binding to serum albumin and slows renal clearance. These changes extend the circulating half-life from roughly two minutes to about one week in humans.
The sequence incorporates alpha-aminoisobutyric acid at position 8, replacing the alanine found in the natural hormone. This substitution blocks the primary DPP-4 recognition site and contributes most of the enzymatic stability. Albumin binding further protects the peptide and reduces the frequency of administration required to maintain active plasma levels. Because the fatty acid chain increases lipophilicity, the compound is formulated as a solution rather than a simple aqueous buffer. Researchers describe the design as an incremental optimization of earlier GLP-1 analogs rather than a wholly new scaffold.
Certificate of analysis documents from suppliers typically report purity by chromatographic area, water content, and counter-ion identity. Independent verification is advisable because reported values can be generated under differing conditions. Impurity profiles matter for research use, where aggregates, deamidation products, and residual solvents may influence experimental results. Container, lot, and chain-of-custody records support traceability. Analytical results are method-dependent, so comparisons between laboratories require the same procedure and reference standards.
Lyophilized peptide material is typically stored at or below -20 °C, with -80 °C used for longer-term archives. Vials should remain sealed and desiccated because moisture promotes aggregation and hydrolysis. Repeated freeze-thaw cycles are avoided since they can alter peptide conformation and reduce recovery. Once reconstituted, solutions are generally kept at 2-8 °C and used within a defined window. Stability beyond those windows depends on buffer composition and concentration, and exact limits are product-specific rather than universal.
Identity and purity are assessed with reversed-phase high-performance liquid chromatography, which separates the peptide from related impurities by hydrophobicity. Mass spectrometry confirms molecular weight and detects truncation or modification products. Peptide mapping after enzymatic digestion verifies the amino acid sequence. Quantitation is often performed by LC-MS/MS or by immunoassay, and the two approaches can give different values because they measure different things. Method validation parameters such as accuracy, precision, and limit of quantitation are reported alongside results.
Handling practices center on minimizing contamination and adsorption. Lyophilized peptide tends to accumulate static charge, so weighing is done with antistatic measures and calibrated balances. Reconstitution with appropriate solvent should be gentle, avoiding vigorous vortexing that generates foam and shear. Solutions are typically aliquoted before freezing to reduce repeated temperature cycling. Personal protective equipment and a fume hood are standard for powder handling.
Reconstituted solutions are less stable than the dry powder, and stability depends on concentration, pH, buffer composition, and container material. Low-protein-binding tubes reduce loss of peptide to plastic surfaces. Some researchers add a carrier protein to limit adsorption at low concentrations. The exact shelf life of a given solution is best determined empirically through a stability study rather than assumed from general guidance, because published data cover only a limited set of conditions.
Poppy seeds from Papaver somniferum are an important food item and the source of poppy seed oil, an edible oil that has many uses. The seeds contain very low levels of opiates and the oil extracted from them contains even less. Both the oil and the seed residue also have commercial uses. The poppy press cake as a residue of the oil pressing can be used as fodder for different animals such as cattle. Especially in the time of the molt of poultry and fancy fowls, the cake is nutritive and fits to their special needs. Next to the animal fodder, poppy offers other by-products. For example, the stem of the plant can be used for energy briquettes and pellets to heat. Poppy seeds are used as a food in many cultures. They may be used whole by bakers to decorate their products or milled and mixed with sugar as a sweet filling. They have a creamy and nut-like flavour, and when used with ground coconut, the seeds provide a unique and flavour-rich curry base. They can be dry roasted and ground to be used in wet curry (curry paste) or dry curry.
== Chemistry == Bulevirtide is a 47-amino acid long lipopeptide with the following sequence: CH3(CH2)12CO-Gly-Thr-Asn-Leu-Ser-Val-Pro-Asn-Pro-Leu-Gly-Phe-Phe-Pro-Asp-His-Gln-Leu-Asp-Pro-Ala-Phe-Gly-Ala-Asn-Ser-Asn-Asn-Pro-Asp-Trp-Asp-Phe-Asn-Pro-Asn-Lys-Asp-His-Trp-Pro-Glu-Ala-Asn-Lys-Val-Gly-NH2 (C13H27CO-GTNLSVPNPLGFFPDHQLDPAFGANSNNPDWDFNPNKDHWPEANKVG-NH2)
=== Antimigraine agents === Antimigraine agents such as the triptans like sumatriptan act as agonists of the serotonin 5-HT1B, 5-HT1D, and/or 5-HT1F receptors. Earlier antimigraine agents were the ergoline derivatives and ergot-related drugs such as ergotamine, dihydroergotamine, and methysergide, which act as non-selective serotonin receptor agonists.
That expansion was primarily driven by Joseph William Willmot (previously employed at the Electric & International Telegraph Company) improving Latimer-Clark's invention in 1870 with the "double sluice pneumatic valve", and in 1880 with the "intermediate signaller/quick break switch for pneumatic tubes", which dramatically sped up operations and made it possible for a number of carrier messages to be in the tube at any one time. By 1880 there were over 21 miles (34 km) of tube in London. A tube was laid between the Aberdeen fish market office and the main post office to facilitate the rapid sale of the very perishable commodity. While they are commonly used for small parcels and documents, including cash carriers at banks or supermarkets, in the early 19th century they were proposed for transport of heavy freight. It was once envisaged that networks of massive tubes might be used to transport people.
== Side effects == Side effects of tiagabine are dose-related. The most common side effect of tiagabine is dizziness. Other side effects that have been observed with a rate of statistical significance relative to placebo include asthenia, somnolence, nervousness, memory impairment, tremor, headache, diarrhea, and depression. Adverse effects such as confusion, aphasia, stuttering, and paresthesia (a tingling sensation in the body's extremities, particularly the hands and fingers) may occur at higher dosages of the drug (e.g., over 8 mg/day). Tiagabine has been associated with new-onset seizures and status epilepticus in people without epilepsy in post-marketing surveillance. This may be dose-related, although it has been reported at doses of as low as 4 mg/day, and may also be related to concomitant use of other medications that lower the seizure threshold. Some of these seizures occurred around the time of dose increases. There may be an increased risk of psychosis with tiagabine treatment, although data is mixed and inconclusive. Tiagabine can also reportedly interfere with visual color perception. It has not been found to cause psychomotor, cognitive, or memory impairment. Unlike certain other GABAergic drugs like muscimol, gaboxadol, and CI-966, tiagabine has not been associated with hallucinogenic effects.

==== CYP450 ==== Cytochrome P450 is a very large family of haemoproteins (hemoproteins) that are characterized by their enzymatic activity and their role in the metabolism of a large number of drugs. Of the various families that are present in humans, the most interesting in this respect are the 1, 2 and 3, and the most important enzymes are CYP1A2, CYP2C9, CYP2C19, CYP2D6, CYP2E1 and CYP3A4. The majority of the enzymes are also involved in the metabolism of endogenous substances, such as steroids or sex hormones, which is also important should there be interference with these substances. The function of the enzymes can either be stimulated (enzyme induction) or inhibited (enzyme inhibition).
== Epidemiology == With the difficulty in diagnosing a TIA due to its nonspecific symptoms of neurologic dysfunction at presentation and a differential diagnosis including many mimics, the exact incidence of the disease is unclear. It was estimated to have an incidence of approximately 200,000 to 500,000 cases per year in the US in the early 2000s according to the American Heart Association. TIA incidence trends similarly to stroke, such that incidence varies with age, gender, and different race/ethnicity populations. Associated risk factors include age greater than or equal to 60, blood pressure greater than or equal to 140 systolic or 90 diastolic, and comorbid diseases, such as diabetes, hypertension, atherosclerosis, and atrial fibrillation. It is thought that approximately 15 to 30 percent of strokes have a preceding TIA episode associated.
=== Normal–phase chromatography === Normal–phase chromatography was one of the first kinds of HPLC that chemists developed, but has decreased in use over the last decades. Also known as normal-phase HPLC (NP-HPLC), this method separates analytes based on their affinity for a polar stationary surface such as silica; hence it is based on analyte ability to engage in polar interactions (such as hydrogen-bonding or dipole-dipole type of interactions) with the sorbent surface. NP-HPLC uses a non-polar, non-aqueous mobile phase (e.g., chloroform), and works effectively for separating analytes readily soluble in non-polar solvents. The analyte associates with and is retained by the polar stationary phase. Adsorption strengths increase with increased analyte polarity. The interaction strength depends not only on the functional groups present in the structure of the analyte molecule, but also on steric factors. The effect of steric hindrance on interaction strength allows this method to resolve (separate) structural isomers. The use of more polar solvents in the mobile phase will decrease the retention time of analytes, whereas more hydrophobic solvents tend to induce slower elution (increased retention times). Very polar solvents such as traces of water in the mobile phase tend to adsorb to the solid surface of the stationary phase forming a stationary bound (water) layer which is considered to play an active role in retention.
The 15N HSQC is normally the first heteronuclear spectrum acquired for the assignment of resonances where each amide peak is assigned to a particular residue in the protein. If the protein is folded, the peaks are usually well-dispersed, and most of the individual peaks can be distinguished. If there is a large cluster of severely overlapped peaks around the middle of the spectrum, that would indicate the presence of significant unstructured elements in the protein. In such cases where there are severe overlap of resonances the assignment of resonances in the spectra can be difficult. The assignment of the HSQC spectrum requires other experiments, ideally using triple resonance experiments with 15N and 13C-labelled proteins, that provide sequential connectivities between residues so that the resonances can be linked to particular residues and sequentially assigned. The assignment of the spectrum is essential for a meaningful interpretation of more advanced NMR experiments such as structure determination and relaxation analysis. Chemicals labelled with 15N isotope are relatively inexpensive, and the 15N HSQC is a sensitive experiment whereby a spectrum can be acquired in a relatively short time, the 15N HSQC is therefore often used to screen candidates for their suitability for structure determination by NMR, as well as optimization of the sample conditions. The time-consuming process of structure determination is usually not undertaken until a good HSQC spectrum can be obtained.
=== Science and technology === In classical Greece, Aristotle (384–322 BC) commented on their colour-changing abilities, both for camouflage and for signalling, in his Historia animalium: "The octopus ... seeks its prey by so changing its colour as to render it like the colour of the stones adjacent to it; it does so also when alarmed." Aristotle noted that the octopus had a hectocotyl arm and suggested it might be used in reproduction. This claim was widely ignored until the 19th century. It was described in 1829 by the French zoologist Georges Cuvier, who supposed it to be a parasitic worm, naming it as a new species, Hectocotylus octopodis. Other zoologists thought it a spermatophore; the German zoologist Heinrich Müller believed it was "designed" to detach during copulation. In 1856, the Danish zoologist Japetus Steenstrup demonstrated that it is used to transfer sperm, and only rarely detaches.

Native GLP-1 is degraded within minutes by dipeptidyl peptidase-4 and cleared quickly. Semaglutide carries a position 8 substitution that blocks that cleavage and a fatty diacid chain that binds albumin. Together these changes extend its circulating half-life to about one week.
Yes. Stimulation of insulin secretion is glucose-dependent, meaning the effect is larger when blood glucose is high and minimal when it is normal. This property separates GLP-1 receptor agonists from agents that drive insulin release regardless of glucose level.
Receptor expression in hypothalamic and brainstem regions is well documented, and reduced energy intake is consistently observed. The relative contribution of central versus peripheral signaling to total weight change is still an open question addressed by ongoing research.
It is given either as a once-weekly subcutaneous injection or as an oral tablet taken once daily. The two forms use different absorption strategies, so they are not interchangeable on a milligram-for-milligram basis.
